چکیده

Squamous cell carcinoma is the main histological subtype of esophageal cancers in China. Clinically, cisplatin-based chemotherapy is the main approach for many patients with advanced esophageal cancer. Most patients with advanced esophageal cancer experienced chemotherapy failure, however, mechanisms for chemotherapeutic resistance have not been completely understood (Bazarbashi et al., 2008; Cho et al., 2008; Takashima et al., 2008). The transcription factor hypoxia inducible factor-1 (HIF-1), one heterodimer composed of HIF-1α and HIF-1β, is a critical mediator of the response to hypoxia. Recent studies revealed that hypoxia was associated with poor responses to chemotherapy in esophageal cancers (Koukourakis et al., 2001). More recently, evidences showed that increased expression of HIF-1α was closely correlated with the growth, invasion and resistance to chemotherapeutic agents of human cancer cells including ESCC. In contrast, the loss of HIF-

Effects of siRNA-mediated HIF-1α gene silencing on angiogenesis in osteosarcoma

OBJECTIVE To explore angiogenesis in osteosarcoma under the condition of hypoxia-inducible factor (HIF)-1α gene silenced by small interference RNA (siRNA). METHODS The SaOS-2 osteosarcoma cells, transfected with the recombinant plasmid pSilencer2.1-HIF-1α or pSilencer2.1-SCR, were classified as HIF-1α/siRNA group or SCR/siRNA group, respectively. In which, vascular endothelial growth factor (...

Inhibition of hypoxia inducible factor 1α expression suppresses the progression of esophageal squamous cell carcinoma.

Hypoxia inducible factor 1α (HIF-1α) is highly expressed and is implicated in the progression of esophageal squamous cell carcinoma. To investigate the potential mechanism by which HIF-1α contributes to the progression of esophageal squamous cell carcinoma, here we established stable esophageal carcinoma cell lines Eca-109 and TE- 13 in which HIF-1α was depleted by shRNA mediated gene silencing...

Enhanced renoprotective effect of HIF-1α modified human adipose-derived stem cells on cisplatin-induced acute kidney injury in vivo

Current therapeutic options for acute kidney injury (AKI) are limited to the use of supportive measures and dialysis. A recent approach that has sparked great interest and gained enormous popularity is the implantation of stem cells to repair acutely damaged kidney organ. Hypoxia inducible factor-1α (HIF-1α) is effective in protecting the kidney from ischemia and nephrotoxicity. In this study, ...

Hypoxia-induced increases in A549/CDDP cell drug resistance are reversed by RNA interference of HIF-1α expression.

This study aimed to investigate the effects of knocking down hypoxia-inducible factor-1α (HIF-1α) through RNA interference on hypoxia-induced increases in drug resistance in A549/CDDP cells, and to study the underlying mechanisms. A small interfering RNA (siRNA) eukaryotic expression vector targeting HIF-1α was constructed and transfected into A54...

Tumorigenicity of Esophageal Cancer Stem Cells (ECSCs) in nude mouse xenograft model

Background and objectives: Modeling cancer in vivo is a very important tool to investigate cancer pathogenesis and molecular mechanisms involved in cancer progression. Laboratory mice are the most common animal used for rebuilding human cancer in vivo. Cancer stem cells (CSCs) are the main reason of failure in cancer therapy because of tumor relapse and metastasis. Isolation of cancer stem cell...
